Interpretive Summary:

Technical Abstract: Previous analyses of the Mycosphaerella graminicola DNA fingerprint probe pSTL70 revealed that it contains a partial reverse transcriptase (RT) gene and three 29-bp direct repeats, which are characteristics of transposons. The DNA sequences of five clones that hybridized with pSTL70 showed that clones 11E8 and 1D5 contain a partial sequence of the RT gene and clones 1D5 and 9A5 contain five and six copies, respectively, of the 29-bp direct repeats. The complete sequence of the 4.1 kb fragment in clone 1D5 contains a partial RT gene, but the flanking region of the gene has no similarity to any sequence in GenBank. The coding region of this RT gene contains many stop condons. Therefore, this clone may not be an active copy of the transposon and may have been inactivated by the fungus. We used inverse PCR to amplify the coding region of this RT gene and extended the sequence by about 300 bp but still did not obtain the complete sequence. Combining the two parts of this gene gave a higher E-value in a BLAST search, which increases our confidence that there is a transposon in probe pSTL70. We currently are making a cosmid library to obtain the complete sequence of this transposable element.
