Title: Chemoenzymatic Preparation of Novel Substrates for Feruloyl Esterases

Interpretive Summary: The enzyme FAE helps break down certain types of chemical bonds in plant cell walls. This process is important in digestion of forage plants by ruminants such as sheep and cattle. The enzyme is also of interest in the modification of plant matter for extraction of natural products. This paper describes two new types of compounds that will be very useful in analyzing the activity of FAE enzymes. A key step in making the new compounds involves use of a biocatalyst, rather than harsh chemical methods. This research provides a new tool for scientists studying this important enzyme.

Technical Abstract: Two novel substrates of feruloyl esterase, 4-nitrophenyl and 2,2,2-tri- fluoroethyl esters of 4-hydroxy-3-methoxycinnamic acid, were prepared from ferulic acid in a four step procedure including chemoselective enzymatic deprotection of acetylated compounds by Lipase PS-30.
