Interpretive Summary:

Technical Abstract: Studies have documented that the genes resistant to different plant pathogens, fungi, bacteria, viruses and nematodes, are highly conserved in some motifs among a variety of plant species. Based on these studies, the disease resistance (R) genes have been isolated from several important plant species. To tag, clone and identify different disease resistance genes in cotton, we have designed a pairs of degenerate primers, based on the conserved P Loop and membrane spanning motif sequences of several genes resistant to the above four pathogens cloned from a variety of plant species. The genomic DNA from cotton line Auburn 634, which is highly resistant to root-knot nematode, was screened with this pair of primers. Two bands were isolated and one of them was cloned. One hundred random clones were sequenced and eighty of them have readable sequences. Sequence analysis showed that twenty-three clones have a similarity of 61 to 90% to the cloned resistance genes from different plant species at nucleotide level. These clones have been mapped to a genetic map using the TM-1 x 3-79 segregation population. Characterization and mapping of these R genes against different cotton diseases, nematodes, fungi, bacteria and viruses, is in progress.
