Interpretive Summary:

Technical Abstract: Interleukin-13 is an anti-inflammatory mediator produced by T cells that also stimulates the differentiation of B cells. We sought to identify single nucleotide polymorphisms (SNPs) in the bIL-13 gene to eventually determine if specific haplotypes were associated with improved or diminished IL-13-associated immune responsiveness. Candidate SNPs were identified by sequencing PCR amplification products of the bIL-13 gene flanking and containing exon 1. Four families in the MARC reference population were initially screened to identify polymorphisms, which were then verified by segregation in the entire reference population using matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Additionally, polymorphisms were identified in 96 individuals representing 17 cattle breeds popular in commericial cattle operations in the United States. Identification of SNP haplotypes for bIL-13 contributes to our pool of SNP markers for immune response gene in cattle, will assist in the design of studies to determine the contribution of allelic variation to immunity, and provides additional information on the genetic make-up of commercial populations which can be used to select livestock superior in immune responsiveness.
