Interpretive Summary: Pig chromosome 8 appears to contain genes which affect reproduction. To facilitate the identification of these genes, we developed new genetic markers specifically targeting chromosome 8. The results of this effort yielded 14 new markers for chromosome 8, 10 markers for chromosome 15, and one marker each for chromosomes 5 and 10. The markers reported from this study will be useful in the identification of genes affecting reproduction on pig chromosome 8.
Technical Abstract: Chromosome 8 is a priority chromosome in efforts to map the pig genome because it appears to harbor some QTL that favorably impact reproductive efficiency. A microsatellite enriched microdissected chromosome 8 library was developed and screened. The microsatellite primers were developed and used to genotype the MARC swine reference population. Two-point linkage analyses were conducted between these markers and all markers present in the MARC genome database. We isolated 14 informative microsatellite markers which mapped to chromosome 8. In addition, 10 informative microsatellites mapped to chromosome 15, one to chromosome 10, and one to chromosome 5. The most likely explanation for markers located on chromosomes other than 8 is that these chromosomes were inadvertently contacted by the microdissection needle, thus contaminating the sample. These additional markers, while not mapping to the desired region, are still valuable since they add informative markers to the genetic map in other regions of the genome. The markers reported in this study may be useful for scientists to map QTL on porcine chromosome 8, 15, 10 or 5.
