Interpretive Summary:

Technical Abstract: A high-resolution maize molecular map is being produced using RFLP and SSR markers. The mapping population is derived from a cross of B73 x Mo17 which underwent random mating for 4 generations. The random mating process increased the efficiency of mapping closely linked markers by 3-fold compared with F2- or RI-derived mapping lines. The map presented here represents the initial phase of a large-scale mapping effort whose goal is to produce a map containing 1000 RFLP and 2000 SSR markers. The RFLP markers have been preselected for low copy number based on prior map information. The loci they identify will serve as a bridge to prior map information. In addition, the 1000 RFLP probes will be mapped against a maize BAC library with 10X coverage providing anchor points for BAC contig assembly based on fingerprinting data. The SSR markers include both prior, publicly-available markers and novel markers derived from new public sequence data and sequencing of enriched libraries. As with the RFLP probes, the SSR markers will provide a bridge from this map to other SSR maps. The SSR markers will also be used to anchor the BAC libraries using "overgoes" produced from flanking 3' sequence. All map data and associated marker information will be available through the Maize Genome Database or via links from the Missouri Maize Project web site.
