Title: Isolation and Evaluation of Soy Isoflavonoids Acting As Chemoprotectants in Mammalian Cell Culture Systems

Technical Abstract: Previous work has shown that a number of compounds isolated from commercial soybean processing by-products have the ability to repress induced genomic DNA damage, whole cell clastogenicity, and point mutation in mammalian cells. Further evaluation of the activity of genistin, genistein, daidzin, and daidzein in the single cell gel electrophoresis (SCGE) model system using CHO AS52 cells demonstrated that the isoflavones expressed variable effects. Genistin repressed the mutagenic activity of 2-acetoxyacetylaminofluorene (2AAAF), while genistein, the aglycone, enhanced the DNA damage induced by 2AAAF. In a separate assay, genistein strongly suppressed the growth rate of human colon cancer cells (HT-29). In order to pursue these studies, larger quantities of chemically defined materials and purified isoflavones are needed. New methodology is being developed to isolate large quantities of purified isoflavonoids from soy products and to rapidly quantitate samples for their isoflavone content. This will allow for more rapid evaluation of samples in bioassay systems and in animal and human studies.
