Interpretive Summary: Charcoal rot is a disease that is a world-wide problem in soybean production for which no highly resistant cultivars are currently available. Soybean germplasm line DT99-16864, a maturity group V line, was developed by the U.S. Department of Agriculture-Agricultural Research Service, Stoneville, Mississippi and Jackson, Tennessee. It is a conventional germplasm line with well-documented moderate resistance to charcoal rot disease. It has resistance to southern stem canker and moderate resistance to soybean cyst nematode race 3 populations. It was tested in multiple environments in ARS tests in Mississippi and the Uniform Soybean Tests – Southern States. In the Uniform Soybean Tests over 41 tests at 22 locations in 2003 and 2004, DT99-16864 was not significantly different than the check cultivars '5002T’ and ‘5601T’ for yield. DT99-16864 will be useful as a disease resistant parent for cultivar development and a check line for disease screening.

Technical Abstract: Charcoal rot, caused by Macrophomina phaseolina (Tassi) Goidanich, is a disease that is a world-wide problem in soybean production for which no highly resistant cultivars are currently available. Soybean germplasm line DT99-16864, a maturity group V line, was developed by the U.S. Department of Agriculture-Agricultural Research Service, Stoneville, Mississippi and Jackson, Tennessee. It is a conventional germplasm line with well-documented moderate resistance to charcoal rot disease. It has resistance to southern stem canker (Diaporthe phaseolorum (Cooke & Ellis) Sacc. var meridionalis F. A. Fernández) and moderate resistance to soybean cyst nematode (Heterodera glycines Ichinohe, race 3) populations. DT99-16864 originated from a single F5 plant derived from a cross of ‘S59-60’ x ‘Bolivar’. The breeding population was advanced from the F2 to the F5 generation using the single-seed descent method. Based on colony-forming units, root and stem severity scores and other rating methods, DT99-16864 had significantly lower levels of M. phaseolina infection than susceptible checks in trials in Stoneville, MS and Jackson, TN. It was tested in multiple environments in ARS tests in Mississippi and the Uniform Soybean Tests – Southern States.
