Interpretive Summary: The newly developed Ultra-performance liquid chromatography (UPLC) method is suitable for rapid analysis of seven alkaloids and chemical fingerprint analysis. The method was tested for all the parameters and successfully applied to the identification of seven alkaloids in plant sample and ten dietary supplements that claim to contain goldenseal and Echinacea purpurea/goldenseal combination formulations. The plant material and ten dietary supplements were found to contain major alkaloids, hydrastine and berberine. liquid chromatography(LC)-mass spectrometry was used for the identification of seven alkaloids in plant sample and dietary supplements. The developed UPLC-MS method can be useful in identification of various other analytes present in plant sample.

Technical Abstract: UPLC with UV detection was used for the quantification of alkaloids from roots of Hydrastis canadensis L. (goldenseal) and dietary supplements claiming to contain goldenseal. The chromatographic run time was less than 6 min. The detection wavelengths used were 290 and 344 nm for '-hydrastine, canadine, hydrastinine, coptisine, jatrorrhizine, palmatine and berberine. The samples extracted with the 90 % methanol (1 % acetic acid), displayed the best recovery (>97%). LC-mass spectrometry with a quadrupole mass analyzer and ESI source was used for confirmation of the identity of seven alkaloids.
