Interpretive Summary:

Technical Abstract: The formation of infection cushions by Rhizoctonia solani (isolate G-24) and R. cerealis (isolate Fellers) was examined on cellophane membranes in response to stimulation by roots of peanut (Okrun, Tamspan 90, Southwest runner and Line 209) and hard red winter wheat (Jagger, 2137, and 2174). Root systems were each enclosed in pouches made from 2.7 cm dia. dialysis tubing. Pouches were placed into perlite filled cups infested with mycelial fragments of the fungus and maintained in a lighted humidity chamber at 22 degree C. After four days, eight to ten squares (2 cm (^2)) were cut from each pouch, stained in 0.5% trypan blue and placed on a glass slide. The number of infection cushions was counted in each square at 100X. The number of infection cushions per square (ICS) formed on peanut by G-24 ranged from 4-8, and no significant (P=0.05) differences were observed among peanut cultivars. Among wheat cultivars infested with Fellers, 2137 had 1 ICS compared to 6 ICS for 2174 and 10 ICS for Jagger. These results suggest that quantification of infection cushions may be a useful technique to evaluate reaction of peanut and wheat germplasm to diseases caused by Rhizoctonia spp.
