Technical Abstract: Cucumber green mottle mosaic virus (CGMMV), first described in England, 1935, is a well-known, seed-borne Tobamovirus on cucurbits in Asia, Europe, and the Middle East. The recent outbreaks of CGMMV in Australia and North America (Canada and U.S.) received great concerns from the vegetable seed companies and cucurbit industries. With the increasing activities in off-shore hybrid seed production and global seed trade, sensitive and reliable seed health assay to CGMMV is imperative to ensure that cucurbit growers are using certified virus-free seed lots. However, the presence of genetic diversity with two distinct genotypes (Asian and European) and recent understanding of serological specificity among CGMMV isolates earned concerns for accurate disease diagnosis using serological tests (ELISA or Lateral flow device). Genetic analysis using the complete genome sequence from Canada revealed a strong sequence identity (99%) with numerous Asian isolates, but with only 90% to several European isolates. Genetic diversity was also observed in other CGMMV isolates, i.e., 99% identity between Australia and Asian isolates and 95% identity between California and European isolates. The poor serological reactivity between these two major genotypes made some commercial ELISA kits not reliable for seed health assay. To improve sensitivity and reliability for broad spectrum detection of CGMMV isolates in all genotypes, a loop-mediated isothermal amplification (LAMP) was developed and used to monitor CGMMV infection on greenhouse cucumber. This simple assay would provide a timely and reliable detection of CGMMV in plant health and seed health tests.
