Interpretive Summary:

Technical Abstract: Proteins play an important role in several biological processes. Proteomics encompasses basically four principal applications, namely protein mining, protein expression profiling, protein-network mapping and mapping of protein modifications. The results in these applications depend mostly on the choice of sample preparation for protein extraction. Two major challenges of protein extraction in proteomics are contaminants and masking abilities of the highly abundant proteins in recalcitrant tissues. In this chapter, methods of contaminant removal such as TCA/acetone, NH4AC/methanol, and phenol/chloroform are discussed along with methods of abundant protein depletion including precipitation/fractionation, antibody based affinity, and filter aided separation. In addition, we provide some perspective of sample preparation strategies for further research related to proteomics.
