Technical Abstract: Biolistic transformation of cotton meristems, isolated from mature seed is detailed in this book chapter. This method is simple and avoids the necessity to use genotype-dependent regenerable cell cultures. However, identification of germ line transformation using this method is laborious and time-consuming. The detailed protocol on meristem transformation is described in this chapter for generating germ line transformation events as well as transformation of superficial layers (epidermis) of plant organs derived from bombarded cotton meristem. In addition, this procedure offers a quick method to evaluate gene constructs in cotton cells, organs, and tissues especially fibers which originate as single cells from the maternal epidermis layer of cotton ovules. This procedure will be useful to students, technicians, and scientists interested in cotton biotechnology.
