Interpretive Summary: TifGP-2 is a runner-type peanut germplasm line that was released by the USDA-ARS and the Georgia Agricultural Experiment Station in 2010. TifGG-2 is a late generation derived sister line of Tifguard, a peanut variety that has high resistance to both the peanut root-knot nematode and tomato spotted wilt virus (TSWV). These highly related sister lines with and without nematode resistance can be valuable research tools to obtain a better understanding of the interactions of nematodes with other pathogens of peanut. Breeding populations were developed and selection for nematode resistance was conducted using standard greenhouse screening techniques. Selection for TSWV resistance was conducted in the field with natural virus infection. The variety Tifguard was selected that had high resistance to both pathogens. A near isogenic line (TifGP-2) without resistance to the peanut root-knot nematode was also developed. Both breeding lines exhibited higher resistance to TSWV and higher yield than standard check cultivars when grown in fields with little or no nematode pressure. Both isolines are available to the research community, and have proved to be be valuable experimental tools to answer important questions about the interactions of nematodes and other peanut diseases. These sister lines should also be valuable tools for research on resistance mechanisms and genetic control of nematode resistance in peanut.

Technical Abstract: TifGP-2 (PI 663944) is a runner-type peanut (Arachis hypogaea L. subsp. hypogaea var. hypogaea) germplasm line that was released by the USDA-ARS and the Georgia Agricultural Experiment Stations in 2010. It was developed at the University of Georgia Coastal Plain Experiment Station, Tifton, GA, and was tested under the experimental designations C724-19-25. TifGP-2 is a late generation derived sister line of Tifguard, a peanut cultivar that has high resistance to both the peanut root-knot nematode [Meloidogyne arenaria (Neal) Chitwood race 1] and Tomato spotted wilt Tospovirus (TSWV), the cause of tomato spotted wilt. During the coarse of developing Tifguard, we also had the opportunity to develop this highly related sister line without nematode resistance. These late generations sister lines with and without nematode resistance can be valuable research tools to obtain a better understanding of the interactions of nematode resistance with other pathogens of peanut. The original breeding population was developed by hybridizing the TSWV resistant cultivar, C-99R with the nematode resistant cultivar, COAN. Selection for nematode resistance was conducted using standard greenhouse screening techniques. Selection for TSWV resistance was conducted in the field with natural virus infection. A cultivar, Tifguard, was selected that had high resistance to both pathogens. A late generation sister line, TifGP-2, susceptible to the peanut root-knot nematode was also selected. These sister lines have proved to be valuable research tools to evaluate the interaction of the peanut root-knot nematode with other economically significant pathogens of peanut.
