Technical Abstract: Structural analysis of carbohydrates involves three parameters: composition, linkage, and conformation, and tends to rely on the various forms of two techniques; mass spectrometry (MS) and nuclear magnetic resonance (NMR) spectroscopy. These techniques are enhanced and extended by the use of stable isotopes, especially **13C and **2H. Because these isotopes can be selectively detected they are typically used in metabolic pulse-chase experiments similar to radiolabels. This presentation will focus on three newer aspects: 1. GC-MS analysis of global metabolic labeling from **13C precursor sugars; 2. Hydrogen-deuterium exchange mass spectrometry (HX-MS) for oligosaccharide conformational studies; and 3. Stable isotope-enhance diffusion-ordered **13C-NMR spectroscopy (SIE-DOSY **13C-NMR). Applications and advantages of these techniques for the study of microbial carbohydrate metabolism will be discussed.
