Technical Abstract: At two days post treatment, a CpG oligodeoxynucleotide (CpG 120818-9A) offered Nile tilapia (Oreochromis niloticus L.) significant (P<0.05) protection against Streptococcus iniae infection, with relative percent survival up to 63%. To understand the molecular mechanisms involved in the protective immunity elicited by CpG 120818-9A, suppression subtractive cDNA hybridization technique was used to identify upregulated genes in the posterior kidney of Nile tilapia in response to CpG 120818-9A treatment. A total of 69 expressed sequence tags (ESTs) were identified from the subtractive cDNA library. Quantitative PCR revealed that 44 of the 69 ESTs were significantly (P<0.05) upregulated in Nile tilapia by CpG 120818-9A treatment, including 15 moderately upregulated (<10 fold) and 29 highly upregulated (>10 fold) ESTs. Of the 15 moderately upregulated ESTs, EST 11D02 (putative cytochrome c oxidase subunit II) was upregulated the highest. Of the 29 highly upregulated ESTs, EST 12F06 (putative peroxisomal sarcosine oxidase) was upregulated the highest, followed by EST 11B02 (putative glutaredoxin-1).
