Technical Abstract: The association of necrotrophic soil-borne pathogens, Rhizoctonia spp., with a root-attacking weevil, Ceutorhynchus assimilis, and diseased tissues of Lepidium draba, an invasive perennial weed in the U.S., has been documented in its Eurasian native range. Among all isolates of Rhizoctonia solani and Rhizoctonia-like fungi collected across the Eurasian range and for which virulence was evaluated, two isolates from France, E06f-11 and E07f-12, were further characterized using nuclear gene sequencing. The targeted sequences were located on the 18S ribosomal RNA gene (partial sequence); internal transcribed spacer 1, 5.8S ribosomal RNA gene and internal transcribed spacer 2 (complete sequences), and 25S ribosomal RNA gene (partial sequence) (Genbank accession numbers HQ738653 to HQ738654). Both isolates were assigned to binucleate Rhizoctonia using sequencing data and following observation of their nucleus status through microscopy. These two isolates will be used to test the specificity of a future real time PCR assay for monitoring Rhizoctonia solani in insect galled roots of Lepidium draba subsp. draba.
