Interpretive Summary: Marker-assisted selection (MAS) is a process where a marker (frequently molecular marker) is used for indirect selection of desired material. This presentation reviews the current status and perspectives of marker-assisted selection in lettuce. A practical example is provided for mapping a resistance gene against lettuce dieback and development of molecular markers for MAS. Detailed information is given for the use of High-Resolution DNA Melting Analysis (HRM) in lettuce breeding at the USDA-ARS in Salinas, CA. HRM is a technique that allows detection of mutations in DNA samples.

Technical Abstract: This presentation reviews the current status and perspectives of marker-assisted selection in lettuce. A practical example is provided for mapping a resistance gene against lettuce dieback and development of molecular markers for marker-assisted selection.
