Interpretive Summary:

Technical Abstract: This poster presentation reports progress on a USDA NRI grant to enhance understanding of and access to the genetic diversity in wild and landrace relatives of tomato, potato, sweet potato, and carrot, and contribute to the same for other euasterid plant species. This is being done with Conserved Orthologous Sequences (COSII sequences), a class of DNA sequences that are potentially orthologous (single-copy), and hence of great utility for applications as diverse as genetic linkage mapping, phylogeny, and diversity. We are sequencing 24 of these markers for phylogenetic studies in potato and tomato, applying these markers to comparative studies of diversity of tomato and potato, and sequencing 420 COS in order to obtain 300 of them for conversion into PCR-based mapping markers. To date we have used 12 COS for well-resolved phylogenies of a core subset of tomato and potato accessions and expanding these now to the full range of 24 COS and more accessions of tomatoes and potatoes. The mapping data are in progress. The data are providing more rational grouping concepts of these crops and providing a useful COSII based map of potato of use to potato breeders.
