Archives of Virology. 154:891–894. Interpretive Summary: Red clover vein mosaic virus (RCVMV) is a serious pathogen of legume pulse crops including pea, chickpea and lentil. RCVMV is a Carlavirus in the family Flexiviridae. Symptoms in these crops include chlorosis, stunting, rosetting, and vein clearing, but early infection also causes plant death in chickpea and lentil. Such symptoms are unusual for a carlavirus, as most other members of the genus cause only minimal symptoms or no symptoms at all. The complete nucleotide sequence was generated from an isolate obtained from chickpea in Washington State. The complete genome of RCVMV was found to consist of 8605 nucleotides and is organized into six open reading frames (ORFs), each coding for a protein in virus-specific functions. The genome characterization shows that it is typical of members of the genus Carlavirus. Phylogenetic analysis showed that RCVMV grouped most closely with Pea streak virus, another carlavirus that causes severe symptoms and losses in pulse crops. RCVMV was most distantly related to Narcissus symptomless virus. This is the first report of the complete nucleotide sequence of RCVMV. Report of ths information will provide for development of better diagnostics strategies and for the study of its epidemiology. Furthermore, it will provide information for studies to determine how this virus evolved to cause such severe symptoms in a viral genus that generally causes only mild symptoms in plants.
Technical Abstract: Red clover vein mosaic virus (RCVMV) is a serious pathogen of legume crops including pea, chickpea and lentil. The complete nucleotide sequence was generated from an isolate obtained from chickpea in Washington State. The complete genome of RCVMV consists of 8605 nucleotides excluding the poly(A) tail and is organized into six open reading frames (ORFs). Amino acid identities and phylogenic analysis using the replicase gene and coat protein gene confirm that RCVMV is a member of the genus Carlavirus in the family Flexiviridae. Phylogeny demonstrated that RCVMV is most closely grouped to carlavirus Pea streak virus. This is the first report of the complete nucleotide sequence of RCVMV. The data will provide for development of rapid and reliable diagnostic strategies as well as for viral evolutionary studies.
