Interpretive Summary: Determination of the genomic make up is fundamental to our understanding of biological systems and can provide a foundation for the development of new vaccines and treatments to prevent and cure disease. A device was invented that shortens the time needed to resuspend cells in one of the first steps in the process of determining the genomic sequence. This device overcame problems in resuspension of cells and increased the efficiency of genomic sequence determination.

Technical Abstract: Genomic sequencing involves sequence determination of inserts from a large numbers of plasmids of a genomic library. In high throughput DNA sequencing projects, cultures are grown in a 96-deep well plate format, centrifuged, and resuspended. Resuspension of cells was aided by use of a simple device that we invented to deliver a single glass bead to each well before vortexing. The device, consisting of a sticky surface applied to a thin, plastic template in a 96-well plate format, is loaded by pouring beads onto the sticky side of the device. Beads are delivered into the wells by inverting the loaded device over the wells so that a bead extends into each well and twisting the device to dislodge the beads into the wells by forcing them against the well's walls. The device was called a Bead-Bop. A modification of the Bead-Bop was made and called the Bead-Bop II. It has a different adhesive and delivered colored beads.
