Objective:
Determine the virus proteins and protein domains regulating tissue tropism of Potato leafroll virus in plant hosts and aphid vectors.

Approach:
Use mutational analysis of three virus proteins previously identified to influence virus movement. This will identify the domains of each of the proteins that are required for observable movement phenotypes. The mutants will also be used to investigate the interaction of the three virus proteins with each other and with host/aphid proteins in various binding and interaction assays as well as using labeled proteins coupled with confocal microscopy.
