Objective:
To conduct association analysis and identify single nucleotide poloymorphism markers associated with bacterial blight resistance through genotyping by sequencing in USDA cowpea germplasm core set and to provide a tool for breeders how to use USDA cowpea resistant resource and to select bacterial blight resistance in cowpea breeding program through marker-assisted selection.

Approach:
Two hundred high resistant and 200 high susceptible cowpea lines from the 720 USDA cowpea germplasm core set have been selected and will be used in this research. Phenotypic data of bacterial blight resistance has been kindly provided by Blair Buckley who will collaborate with Ainong Shi for conducting association analysis of bacterial blight resistance in cowpea. Genotyping of cowpea accessions will be done using genotyping by sequencing. Single nucleotide poloymorphism discovery of GBS data will be done using DNASTAR Lasergene Genomics Suite. The genetic diversity will be analyzed using PowerMarker3.25 and MAGA 5. Genome-wide association studies will be analyzed using TASSEL, CLC Genomics Workbench 6.0 - CLC bio, and SNP & Variation Suite 7 of Golden Helix Inc.
