DETERMINATION AND CHARACTERIZATION OF DEOXYNIVALENOL IN BARLEY

Project Title 1 - Determination of Bound Deoxynivenol in Barley Breeding Lines: Recent reports of bound deoxynivalenol (DON) in barley and wheat are a cause for concern, as by definition, bound DON is that which escapes detection by the routine analytical methods.
The objectives of this one year project are to (1) determine the prevalence of bound DON in commercial samples of barley that have been infected with Fusarium Head Blight (FHB), and (2) to determine how differences in plant resistance to FHB and DON accumulation impact the levels of total and bound DON.
Project Title 2 - Malting Barley Deoxynivalenol Services: The malting and brewing of Fusarium infected barley presents a number of processing, product quality and public health concerns. Fusarium infected barley also is unsuitable for human consumption and for some livestock. The ultimate solution to Fusarium-related problems is the development of FHB resistant barley cultivars. Testing for deoxynivalenol (DON) is an integral part of barley varietal development programs focusing on Fusarium resistance. DON testing, however, is a very expense part of these programs, and thus can limit the number of lines, which may be screened within a given year. The primary objective of this project is to provide barley breeders and pathologists, working on the development of Fusarium resistant barley, with affordable, accurate and timely DON analysis. Funds requested will support the analysis (DON) of up to 16,000 barley samples on an annual basis.
Project Title 1:
Free DON will be determined using our normal procedures of solvent extraction and analysis by GC-ECD or GC-MS. Bound DON will be determined following treatment of samples by TCA solvolysis, or with proteases. The content of DON-3-glucoside will estimated by HPLC following solvent extraction. Samples for objective 1 are to be obtained from a regional barley quality survey, while those for objective 2 will be obtained from the North American Barley Scab Evaluation Nursery (NABSEN). Wheat samples obtained from a regional quality survey, and the NDSU hard red spring wheat variety development trials, will be included for comparative purposes.
Project Title 2:
Analysis (DON) of up to 16,000 barley samples on an annual basis. Analysis of regional barley crop samples will provide annual information on DON levels. An inter-laboratory check service is offered as a means for improving the reproducibility of DON analyses between laboratories.
