Discovering, Understanding, and Utilizing Wheat Genes for Fhb Resistance in Ohio

Project Title 1 - Uniform Nursery for SRWW and Development Scab Resistance Varieties for Ohio: It is our aim to use MAS and phenotypic selection in a program that will insure a steady release of FHB resistant cultivars while building parents for future success.
Project Title 2 - Mapping QTL for Type I and II FHB Resistance from CIMMYT Germplasm derived from a Synthetic Hexaploid: Producing grain with low DON is the ultimate goal for of controlling Fusarium Head Blight, caused by the fungus Fusarium graminearum.
Project Title 3 - Mapping Fusarium Head Blight Resistance in Truman Wheat: Our objective is to use QTL mapping to identify FHB resistance genes from Truman.
Project Title 4 - Mapping FHB QTL in an IL97-1828 x Clark Derived RIL Population: Our objective in this research project is to identify molecular markers associated with loci for FHB resistance in a source that does not trace back to a Chinese source of FHB resistance (so called “native” resistance).
Project Title 5 - The Molecular Basis for the Low DON Accumulation Phenotype in Winter Wheat: We will test three hypotheses in this proposal:
1. Wheat lines with different levels of RDA have different effects on transcription of Fusarium graminearum genes.
2. Wheat lines with different levels of RDA have different effects on translational and post-translational control of DON production and/or degradation.
3. Variation in levels of Mg2+, other minerals, proanthocyanidin, ferulic acid, polyphenol oxidase and catalase activity in wheat embryos or seeds.
Project Title 1 - (1) Evaluate the FHB resistance of all breeding lines in OSU program develop and populations from which FHB resistant cultivars can be developed in the future; (2) Evaluate the FHB resistance of cultivars in the OSU Commercial Cultivar Trial; (3) Backcross FHB resistance QTLs into promising OSU breeding lines in early stages of their evaluation; and (4) Coordinate a uniform nursery for evaluating FHB reaction in SWW adapted to the northern U.S.
Project Title 2 - Identify unique genes for Type I and II FHB resistance from CIMMYT germplasm derived from a synthetic hexaploid.
Project Title 3 - In collaboration with the genotyping center at Raleigh N.C., we have preliminarily haplotyped Truman and determined that the source of resistance in Truman differs from both Chinese germplasm and from Ernie.
Project Title 4 - Use QTL mapping to identify FHB resistance genes from IL97-1828.
Project Title 5 - The hypotheses address the 2nd and 3rd action plan goals of the Pathogen Biology and Genetics research area. We will spray inoculate two resistant and two susceptible lines in the greenhouse. For each line we will collect embryos/developing seeds at 2, 4, 8, and 16 days after inoculation. For each sample from each line we will assay the content of Fusarium graminearum biomass, DON, minerals, ferulic acid, proanthocyanadins, and polyphenoloxidase and catalase activity. We will conduct a micro-array analysis of the expression of fungal genes on one resistant/susceptible pair using embryos sampled at 2 and 4 days.
