Research Project: Eradication Strategies for Globodera pallida, the Pale Cyst Nematode (PCN)

Objective:
1. Develop integrated strategies for the eradication of G. ellingtonae. 2. Determine G. ellingtonae egg decline at Powell Butte.

Approach:
Objective 1 – Several laboratory and greenhouse experiments will be conducted to continue to explore eradication strategies, alone and combined against G. ellingtonae. a. Brassica juncea and Sinapis alba (mustards) seed meals and extracts of these mustard seed meals will be evaluated for ability to induce and/or suppress hatch of G. ellingtonae. The meals and extracts will be applied to soil containing cyst of G. ellingtonae at rates equivalent to 1, 2, 3, and 4 t/a. A nonamended control will be included in the experiment and treatments will be replicated 5 times. The amended soil will be placed into 50 ml tubes, hydrated with water, and placed at room temperature for 1-2 weeks. After the incubation period, cysts will be extracted from soil. b. The application of LT trap crop with mustard seed meals and extracts for the eradication of G. ellingtonae will be tested in a greenhouse experiment. The experimental design will consist of B. juncea and S. alba extracts and meals, and LT alone (3 treatments) or combined for a total of five treatments plus a nontreated control. Mustard meals and extracts will be added to soil containing G. ellingtonae cysts; pots receiving no mea/LT and LT alone will remain nonamended. c. Additional plants that may serve as trap crops for G. ellingtonae, including lupin and quinoa, will be tested. Objective 2 – To determine the natural decline in G. ellingtonae egg populations over time, the field site at Powell Butte will be sampled. During 2012-2015 potato inoculated with G. ellingtonae was planted in different parts of the experimental area. This spatial distribution of different ages of nematodes allows for the evaluation of reduction in egg numbers of 1, 2, 3, and 4-year-old cysts. In each area previously planted to potato, approximately 45 ft wide by 100 ft long, four transects will be established running the length of the area. From each transect 15-20 soil samples will be collected and combined. Sampling will occur in the spring and fall of 2016 and the spring of 2017. The soil will be removed from bags, dried and weighed, and cysts extracted with a USDA cyst extractor; number of eggs in collected cysts will be determined by crushing over a 500-mesh sieve and counting on an inverted microscope. The number of cysts and eggs/g soil will be determined.
