Evaluation of Barley and Malt for Don and Deoxinivalenol-3-Glucoside

Project 1: Malting Barley Deoxynivalenol Diagnostic Services: Provide barley breeders and pathologists, working on the development of Fusarium resistant barley, with affordable, accurate and timely DON analysis.
Project 2: Formation of Deoxynivalenol-3-Glucoside during Malting: (a) determine the development of DON3G during malting in samples that would be found in commercial use; (b) determine if there are genotypic differences in the capacity of the barley UDP glucosyl transferase system to convert DON to DON-3-G; and (c) determine if the presence of DON-3-G interferes in the analyses of DON using methods employed in commercial practice.
Project 1: Funds requested will support the analysis (DON) of 11,700 barley samples/year from nine researchers in four states. The majority of samples will be tested for DON using GC-ECD. A smaller number of samples require a more comprehensive toxin using either GC-MS or LC-MS/MS. Up to 200 samples/year are to be tested for DON-3-glucoside.
Project 2: Samples will be analyzed for DON by GC-ECD according to the method of Tacke and Casper (1996). DON-3-G will measured by liquid chromatography-mass spectrometry (Simsek et al 2012). Some additional work on sample clean-up and concentration may be required to lower the effective LOQ of this method below 0.1 mg DON-3-G /kg grain.
