Objective:
There are (4) objectives to this project: 1) Complete formal preparation of peer-reviewed manuscripts describing the development of the HPCL-esi(+) MS method of analysis of toxic dehydopyrrolizidine alkaloids (DHPAs) and their N-oxides in comfrey-derived dietary supplements, 2) Determine the major DHPA and N-oxide metabolites produced by Petasites spp., 3) Isolate analytical standards from commercially-acquired, powdered Petasites sp., and 4) Apply and validate the generically-applicable HPLC-esi(+) MS-based method for the quantitative analysis of DHPAs and their N-oxides in Petasites spp. and derived products for human use.

Approach:
The data will be acquired in the closing months of the current project. Data processing, analysis and formal reporting will follow as time permits. Phase II will be a combination of literature researching and empiracal research using the generic HPLC-esi(+) MS method to analyze the DHPA (and N-oxide) content of the commercially obtained Petasistes spp. Phase III will include large quantities of powdered Petasites spp being extracted and the processing of DHPAs. Phase IV will include the initial application of the generic HPLC-esi(+) MS analytical method to determine the need for modifications and to enhance the resolution and detection of the analytes. This method will be validated and applied to commercial Petasites-dervied dietary supplement products.
