1a.Objectives (from AD-416):
1) Identify all electrical penetration graph (EPG) waveforms for adult lygus bugs feeding on susceptible cotton squares, and. 2)Define the biological meanings of waveforms by correlating them with stylet tip locations and stylet activities, especially ingestion from a certain cell type.
1b.Approach (from AD-416):
A Lygus hesperus colony will be established in the laboratory. Thereafter, three studies will be performed, to. 1)identify L. hesperus EPG waveforms,. 2)begin to correlate each waveform with stylet locations in cotton square tissue via cutting stylets and using histological examination to identify cell types penetrated, and. 3)begin to correlate waveforms with stylet activities via simultaneous video-recording and EPG-recording of stylet movements in artificial diet.
This research is in support of objective 3 of the in-house project (determine biological/ecological/transmission properties of Xylella fastidiosa [Xf]), by evaluating feeding of a non-Xf-vector pest species (Lygus hesperus). The cooperator (University of California, Davis) is collecting and rearing Lygus bugs needed for experiments.
