To 1) evaluate the influence that Beet Soil Borne Mosaic Virus (BSBMV) has on aggressiveness and pathogenicity factors of Beet Necrotic Yellow Vein Virus (BNYVV), the virus that causes Rhizomania, which will help predict whether BSBMV affects resistance durability against Rhizomania; 2) determine if sugarbeet genotype influences mutation rate and cross-protection of BNYVV when co-infected with BSBMV; 3) compare different BNYVV isolates for their ability to break-down resistance mediated by the Rz2 gene; and 4) determine the effect that sugarbeet cultivar rotation has on the development of resistance-breaking strains of BNYVV.
Improved understanding of the effect of sugarbeet cultivar and BSBMV have on Rhizomania will be established in greenhouse and laboratory experiments. For all experiments, cultivation of plants under standard greenhouse conditions in artificially infested soil will be performed. Plants will be harvested after several weeks. Virus titer and virus variability in lateral roots will be determined by means of ELISA, RNA extraction, and RT-PCR. Next generation sequencing will be used to identify mutations in pathogenicity genes.
