Lab colonies of psyllids other than potato psyllid, aphids, and leafhoppers will be established from field-collected insects. The insects will be assayed for the ability to transmit Candidatus Liberibacter solanacearum (Lso) following exposure to infected plants. The insects will then be dissected and their tissues will be hybridized with fluorescent probes which bind specifically to Lso. Tissue-specific infection by Lso will be observed using fluorescence microscopy. In addition, seed lots of several potato varieties of export importance will be infected with different haplotypes of Lso, monitored for emergence and Lso infection in above ground and tubers. Furthermore, it will be determined whether potato psyllids can acquire Lso from these plants.
