Research Project: Genotypic Evaluation and Marker Selection of Lolium and Festuca Germplasm

Objective:
Screen a series plant germplasm for the presence/absence of a series of Lolium SSR markers. Germplasm that will be evaluated in the project are IL1, IL2, IL x Festuca hybrids and Festuca dihaploid recoveries.

Approach:
Lolium multiflorum and L. perenne SSR markers will be evaluated across a series of Lolium, Festuca and Lolium x Festuca hybrids; as well as their dihaploid offspring. Initial analysis will follow standard PCR methodology and evaluations will be performed on horizontal based agarose gel electorophoresis. Exceptional markers then will be re-evaluated on a capillary electrophoresis apparatus to determine their validity and obtain a more accurate base-pair size reference. Germplasm to be utilized in the study will be provided by the USDA-ARS. Seed and or plant material will be transferred to cooperators at Cameron University for PCR and marker evaluation
