Determine the characteristics of spontaneous avian leukosis-like tumors in chickens.
1b.Approach (from AD-416):
A pyrosequencing assay will be used to assess the TVB genotypes of commercial broiler breeder chickens (200 chickens) in order to determine the genetic susceptibility of the commercial chickens to ALV-E. Using gene expression analysis by microarrays, and by using DNA Southern blotting and hybridization, the tumors induced in chickens (20 samples) will be characterized and the oncogene involved will be determined.
This project is directly linked to project 3635-32000-016-01C Titled “Etiology and Molecular and Biological Characteristics of Spontaneous Avian Leukosis-Like Tumors in Broiler Breeder Chickens.” A field strain of subgroup E avian leukosis virus (ALV-E) has been propagated, and cloned for sequencing. Spontaneous ALV-like tumors obtained from chickens of USDA-ARS-ADOL line RFS that had been vaccinated with strain SB-1 of serotype 2 Marek’s disease virus and inoculated with ALV-E are being processed for molecular analysis using RNAseq.
