Research Project: Further Studies of Spontaneous Avian Leukokis-Like Tumors in Chickens

Location: Avian Disease and Oncology Laboratory

2012 Annual Report

1a.Objectives (from AD-416):
Determine the characteristics of spontaneous avian leukosis-like tumors in chickens.

1b.Approach (from AD-416):
A pyrosequencing assay will be used to assess the TVB genotypes of commercial broiler breeder chickens (200 chickens) in order to determine the genetic susceptibility of the commercial chickens to ALV-E. Using gene expression analysis by microarrays, and by using DNA Southern blotting and hybridization, the tumors induced in chickens (20 samples) will be characterized and the oncogene involved will be determined.

3.Progress Report:

This project is directly linked to project 3635-32000-016-01T Titled “Etiology and Molecular and Biological Characteristics of Spontaneous Avian Leukosis-Like Tumors in Broiler Breeder Chickens.” In order to determine the genetic susceptibility of the commercial chicken lines to subgroup E avian leukosis virus (ALV-E), the cooperator selected and hired a part-time Technical Support Person to assist in conducting the pyrosequencing assay on blood DNA collected from 200 commercial broiler breeder chickens. The samples are being processed for pyrosequencing assay to determine the TVB genotypes in the commercial chicken lines. The TVB genotypes in lines expressing spontaneous tumors will be compared with those in lines that lack such tumors.
