To increase the genomic data for peanut (Arachis hypogaea) in relation to U.S. production.
1b.Approach (from AD-416):
Genomic data will be generated in two distinct areas. . 1) Reduced representative DNA libraries will be sequenced using a high throughput DNA sequencer. As peanut is a tetraploid with a large genome size it is still too expensive to do whole genome resequencing. This method selectively reduces the areas sequenced but provides useful coverage over many gene regions and allows for the discovery of single nucleotide polymorphisms (SNP) which are useful for genetic mapping. . 2) Gene expression data will be generated using a technique called RNA-Seq. Peanut tissue with spatial and temporal differences will be examined.
The goal of this project is to develop genomics resources for the international peanut genome initiative, specifically in relation to understanding how genes are expressed in various tissue of the peanut plant. Using high-throughput DNA sequencing, Ribonucleic acid (RNA) from various peanut tissues are being deeply sequenced in a replicated experiment. To date eight tissues have been processed though this procedure and more tissues will be processed in next quarter. Once the data has been collected, extensive bioinformatic analysis will be conducted to discover genes important for specific peanut traits and the data will be used in the annotation of the peanut genome when it is developed in 2013/2014. This work is being conducted in conjunction with the University of Georgia (Athens and Tifton campuses).
