1a.Objectives (from AD-416):
To study feeding of glassy-winged sharpshooter.


1b.Approach (from AD-416):
Allow glassy-winged sharpshooters to feed on artificial diet containing an x-ray contrast agent, then video x-ray the feeding at the Argonne National Lab.


3.Progress Report:

Results of this study are in support of Objective 3 of the parent project. Glassy-winged sharpshooters, host plants, and supplies were transported under permit to the Advanced Photon Source at Argonne National Laboratory in April 2011. Sharpshooters were simultaneously videotaped under x-ray phase contrast microscopy and electronically recorded, on both plants and artificial diets. Previous problems with equipment and software were solved. Addition of contrast agents such as iodine and microscopic air bubbles to both plants and diets caused rejection and lack of feeding. However, on a few occasions, entry of air into plants and stylets provided sufficient contrast to partially visualize air and fluid movement into the stylets. Results also showed that sharpshooter feeding does not cause formation of air embolisms in xylem vessels, as had been proposed by some researchers. Future research at Argonne will be needed to expand the sample size for the air embolism study, sufficient for publication.
