The objective of this research is to identify bacterial genes required for tick colonization.
1b.Approach (from AD-416):
A negative-selection screen will be done using a fully characterized, Francisella novicida, two-allele mutant library.
This work relates to objective 1 of parent project by provision of pathogen genome data concerning genes required for efficient transmission. The long-term goal of this project is to identify genes that are required for tick transmission by doing negative selection screens using a mutant library from the tick transmitted bacteria, Francisella novicida. Approximately 20% of the library has been tested in a primary screen. Mutants that were negatively selected during the primary screen have deletions in genes involved in virulence, nutrient acquisition, motility, and DNA repair and recombination. A secondary screen is ongoing to verify the result from the primary screen. Information is exchanged relative to this work by regular face to face meetings and email.
