Research Project: SOYBEAN RAFFINOSE FAMILY OF OLIGOSACCHARIDES ENVIRONMENTAL STABILITY STUDY

Location: Plant Genetics Research

2012 Annual Report

1a.Objectives (from AD-416):
Conduct environmental stability study on soybean genotypes containing distinct oligosaccharide profiles.

1b.Approach (from AD-416):
Selected soybean genotypes have been developed with characteristic oligosaccharide profiles. Seeds from those genotypes will be increased and subjected to an environmental stability study conducted at six locations in the state of Missouri. Harvested seeds will be phenotyped and an HPLC analysis performed for the correlations of environmental factors with differences in oligosaccharide profiles.

3.Progress Report:

This project was complete as of April 1, 2012. The project was a one year environmental stability study for soybean lines with altered raffinose family of oligosaccharides (RFO) profiles. The objective links to the in-house project (to develop the molecular basis for modification of the fatty acid components of soybean oil and anti-nutritional components in soybean meal to use in accelerated breeding programs) by evaluating the environmental impact on the altered oligosaccharide profile trait that can enhance soybean meal value and is controlled by one or more genes. The seeds from five different lines were planted in two experiments: a planting date study and a location study. The measurable outcomes of this project were achieved:.
1)Quantitation of the mean and variation in the galactinol, sucrose, raffinose, and stachyose levels in five distinct soybean genotypes under different environments produced in replicated field trials..
2)Correlations of the RFO values for each of the five genotypes with environmental components such as latitude, planting date, high and low temperature during seed maturation, protein content, and yield.
