Research Project: CHRONIC STAPHYLOCOCCAL MASTITIS: ERADICATION OF PATHOGENS THAT RESIDE INTRACELLULARLY

1a.Objectives (from AD-416)
Test the ability of a fusion protein [lysostaphin fusion to the HIV TAT protein transduction domain] to cure chronic mastitis in cattle.


1b.Approach (from AD-416)
Field trial with chronically infected cows.


3.Progress Report

Two unique staphylolytic fusion antimicrobials harboring three lytic domains have been created and are each being fused to 11 different protein transduction domains. Protein transduction domains allow the extracellular enzymes to be transduced across the cell membrane. These will be compared to the parental lysin for their enhanced ability to eradicate intracellularly localized S. aureus. This work will help to create antimicrobials to cure methicillin-resistant Staphylococcus aureus (MRSA) and other drug resistant staphylococcal infections. Activity on this project is monitored through conference calls.
