1a.Objectives (from AD-416):
This project will establish collaborative research with two institutions of the Indian Council of Agricultural Research working on FMD to conduct antigenic and genetic characterization of FMD field virus isolates, surveillance, monitoring and vaccine strain selection for FMD control programs in India.
1b.Approach (from AD-416):
Researchers from ARS, the Indian Veterinary Research Institute, and the Project Directorate on Foot and Mouth Disease (PDFMD) will:
i) genetically and serologically characterize FMDV strains from cattle, buffalo and sheep circulating in endemic areas of India;
ii) utilize phylogenetic and serological studies to determine the serological relationship of FMDV strains and identify viral strains conferring cross reaction against a wide range of subtypes within FMD serotypes; and
iii) utilize the hAd5-vector platform to generate candidate vaccines containing the P1 region of the Indian vaccine strains (Serotypes O,A and Asia1) and any other widely reactive Indian field strains and test in pilot proof of concept experiment the serological response of animals vaccinated with the novel hAd5-FMD vaccines compared to current vaccines strains.
The overall objective of this project is to identify molecular determinants responsible for antigenic variation of Indian Foot-and- Mouth Disease Virus (FMDV) field strains. This will be accomplished through the development and evaluation of human adenovirus 5 (hAd5)-vector based vaccine against Indian strains of Foot-and-mouth disease viruses and the characterization of cell mediated immune response of Indian cattle and buffalo immunized with hAd5-vector based Foot-and-mouth disease candidate vaccines.
The major accomplishments of FY 2012 include: Making recombinant hAd5-FMDV constructs against three Indian FMD vaccine (O, A and Asia-1) strains and two type A field strains, obtaining an import permit from Department of Biotechnology, Government of India to import the recombinant hAd5-FMDV constructs from PIADC. Also during this year, serotype A FMDV strains from India were phylogenetically analyzed and compared to type A vaccine strain. Amino-acid sequence of strains with low and high r-values were compared to the vaccine strains. Mutations were introduced in the capsid sequence of the vaccine strain to produce recombinant Ad5-FMD vaccines with modified capsid sequence and determine the effect of these modifications on the r-values. Material transfer agreements have been signed to prepare shipment of plasmids and hAd5-FMD vaccines produced during the 6 month training visit of collaborating scientists from India to ARS, PIADC (FY 2011-FY 2012).
The major accomplishments over the life of this project include the construction of FMDV capsid protein expressing recombinant hAd5 viruses against three Indian vaccine (O, A and Asia-1) strains and two type A field strains and the phylogenetic analysis of Indian type A field strains in comparison with vaccine strain and introduction of mutations in the capsid of field strain with low r-value to produce recombinant Ad5-FMD vaccines with modified capsid sequence and determine the effect of these modifications on the r-values.
