1a.Objectives (from AD-416)
1) Develop genetic crosses of selected isolates of P. graminis, P. recondita, P. triticina and P. triticina/P. persistens;.
2)make collections of rust fungi in Israel;.
3)characterize proteins involved in mating in rust fungi; and.
4)characterize spore types and biology of selected rust fungi.
1b.Approach (from AD-416)
Telia of selected isolates will be used to infect appropriate alternate host and genetic crosses will be made from resulting pycnia. Pure lines resulting from the crosses will be developed and characterized on selected host plants. Collections of rust fungi will be made from selected sites in Israel. Biological material will be produced and collected from infection of appropriate hosts. Experiments will be performed to test selected inhibitors on early steps in mating. Spore morphology and life-cycle biology will be characterized for selected rust samples.
Trap nurseries were established at three locations in Israel for monitoring wheat rust pathogens. Collections of Puccinia graminis (wheat stem rust pathogen) were made and characterized. Crosses were performed to develop materials for genetic analysis of P. graminis and P. triticina (wheat leaf rust pathogen). In addition biological materials were generated for genomic analysis. A collection of a leaf rust pathogen from wild durum wheat was characterized and determined to be a new species of rust fungus. Work continued on characterization of rust pathogens of Allium species.
Regular e-mails and an annual visit were used to monitor progress of this project.
