Research Project: GLOBAL GENE EXPRESSION ANALYSIS TO INVESTIGATE MUCOSAL IMMUNE RESPONSE TO AVIAN INFLUENZA IN KOREAN NATIVE CHICKEN BREEDS

Location: Animal Parasitic Diseases

2009 Annual Report

1a.Objectives (from AD-416)
Thorough investigation of the immunogenetic responses involved in host-pathogen interactions in Avian Influenza will lead to better understanding of genetic countrol of host response to AI and the identification of immunogenetic mechanisms which will facilitate the development of novel vaccination strategies against AI.

1b.Approach (from AD-416)
APDL plans to apply avian cDNA intestinal IEL microarray (AVIELA) chips which were developed in Dr. Lillehoj's laboratory at BARC, ARS to characterize the differential immune response patterns of different genetic breeds of broiler chickens with different disease phenotypes.

3.Progress Report

The objective of this grant was to investigate host cytokine response to H9N2 Low Pathogenic Avian Influenza Virus (A/chicken/Korea/MS96/1996(MS96) on three different chicken strains, Ogol (Korean native chicken), Bovans White and Cobb 500. Two-week-old chickens were inoculated with 100 µl of 106.7EID50 by intranasal and oral routes. Lung and spleen of infected chickens were collected on 1, 3 and 5 days after virus infection. Fecal samples were collected on day 5 after virus infection. The results of first trial is being analyzed.

This collaboration was monitored by regular meetings and progress reports.
