2009 Annual Report


1a.Objectives (from AD-416)
The objectives of this cooperative research project are: using genomics tools to 1) identify new QTL for FHB resistance in Asian landraces and U.S. hard winter wheat germplasm from U.S. and.
2)transfer and pyramid QTL for FHB resistance from diverse sources to locally adapted cultivars using marker-assisted selection.


1b.Approach (from AD-416)
Use of high throughput molecular marker technologies to identify and isolate desirable genes/QTL from wheat germplasm materials collected from Asia and the southern Great Plains of the USA and to transfer and pyramid these QTL into adapted U.S. hard winter wheat cultivars using marker-assisted backcross to develop hard winter wheat cultivar/germplasm with improved resistance.


3.Progress Report

Fusarium head blight (FHB) is one of the most important constraints on wheat production and quality in the U.S.A. Use of FHB resistant cultivars is the most effective approach to improve resistance to the disease and lower DON toxin accumulation in wheat grain. The objectives of this cooperative research project are: using genomics tools to.
1)identify new genes for FHB resistance in Asian landraces and U.S. hard winter wheat germplasm lines; and.
2)transfer and pyramid genes for FHB resistance from diverse sources into locally adapted cultivars using marker-assisted selection. A resistance gene (Fhb1) on chromosome 3BS from cultivar Sumai 3 has been transferred into the hard winter wheat cultivars Trego (KS), Wesley (NE), and Harding (SD) using marker-assisted backcrossing. Lines carrying markers were tested in the field FHB nurseries at Manhattan KS and Lincoln, NE. Seeds from Wesley-Fhb1 lines were increased for further agronomic evaluation. Marker-assisted backcross transfer of Fhb1, Fhb2, and resistance genes on chromosomes 4A and 5A were initiated. These genes were transferred into wheat cultivars Jagger (KS), Overley (KS), and Overland (NE). Backcross progeny were selected based on marker data, second backcrosses were made, and seeds were harvested for further marker-assisted selection. Two populations with Chinese/Japanese landraces, Haiyanzhong and Huangfangzhu, as resistant parents have been evaluated in the greenhouse and KSU FHB nursery for FHB resistance. Data will be analyzed by QTL analysis. In addition, a new mapping population Heyne/Trego was developed and was evaluated for FHB in the greenhouse. Progress on this agreement is monitored by regularly discussing program goals, approaches, and results (on site) and by reviewing annual accomplishments reports.
