Objective:
Develop and evaluate infectious recombinant PRRSV clones as potential vaccine candidates. Identify immune targets to differentiate between protected and non-protected animals. Characterization of emerging PRRSV isolates.

Approach:
Infectious modified PRRSV clones will be developed in order to investigate their suitability as vaccine candidates, using in vivo and in vitro studies. Virus will be characterized using DNA sequencing, Northern blot analyses, growth curves, plaque assays, and polyclonal B cell activation (flow cytometry) studies. In vitro testing may include plaque development, viral titers and growth rate analysis, as well as RT-PCR for detection of PRRSV and assays for swine immune responses to the modified virus. In vivo testing will include evaluation of the efficacy of candidate vaccines and other PRRSV constructs in animal tests (swine), including pig growth performance parameters and clinical observations, as well as virus isolation and serological testing.
