Leafy spurge and Canada thistle are perennial weeds that display dormancy and vegetative reproduction from underground adventitious buds. Transcriptomics approaches such as microarray analysis, RNA sequencing, and RT-PCR will be used to determine potential molecular targets that affect vegetative reproduction and dormancy in underground adventitious buds. Data obtained through bioinformatic analysis will be reviewed to identify potential molecular targets with signal transduction pathways that hold the most potential for manipulating dormancy and vegetative reproduction. Likewise, physiological and biochemical methods will be employed to determine to role of plant hormones and growth regulators in dormancy and vegetative reproduction of buds. Experiments aimed at our objectives will be conducted in the laboratory, greenhouse, and field.
