Methods & Techniques
Methods in the Stephensen laboratory focus on evaluation of cellular immune responses in humans and in mice using a variety of methods. Methods include the following:
· Isolation of peripheral blood mononuclear cells or cell subsets (e.g., monocytes, total granulocytes, eosinophils, T-cell subsets) using density gradients, magnetic beads and FACS
· Culturing primary cultures of lymphocytes, monocytes or granulocytes using Luminex or MSD array technology
· Examination of cytokine production using cell culture supernatants or by intracellular cytokine staining and flow cytometric analysis
· Evaluation of vitamin A status using HPLC methods
· Western blot analysis of protein expression
· RNA expression analysis using quantitative RT-PCR or microarray methods
