At the USDA-ARS Plant Genetic Resources Unit (PGRU) we are conserving and genetically characterizing germplasm collections of a diverse array of vegetable crops. Approximately 200 species are conserved in our collection, including : tomato, onion, broccoli, cauliflower, cabbage, asparagus, tomatillo, ground cherry, winter squash, radish, and buckwheat. Typical applications of molecular genetic technology to conservation and management of the germplasm collections involve solving problems in - i) acquisition, maintenance, and genetic characterization of collections; ii) determining levels of genetic diversity and identifying genetic gaps and duplicates in collections; iii) establishing and validating core subsets; iv) monitoring changes in gene frequencies in accessions resulting from regeneration protocols; and v) developing or modifying molecular genetic marker technology for particular crops as needed.
